Bakalářská práce se zabývá stanovením růstu sinic z rybníků. Výsledky slouží ke stanovení růstového potenciálu v souvislosti s eutrofizací, tj. zvyšování koncentrací živin ve vodách. V experimentu byly použity kultivované kmeny Cyanobium gracile A1 a Synechocystis sp. PCC7509. Pro stanovení jejich růstu byla využita statická kultivace v kultivačních destičkách s rozdílnými objemy jamek Thermo Scientific NunclonTMDelta Surface 96 Well a Cellstar 48 Well Cell Culture Plate, které byly z první části pokusu vyhodnocené jako nejvhodnější. Sinice byly kultivovány v koncentracích od 0,06 do 30 mg P/l, v Z médiu. K měření růstu byl využit spektrofotometr, nastavený pro měření fluorescence a absorbance při vlnové délce OD750 nm. Při této vlnové délce mají sinice zanedbatelnou absorbci, a proto se pro měření biomasy využil zákal. Z jednotlivých měření se vytvořily růstové křivky, které jsou v práci zobrazeny pomocí grafů.
Z výsledků lze říci, že nejvyšší použitá koncentrace, 30 mg P/l, způsobila inhibici růstu. Sinice v prostředí o koncentraci 0,06 mg P mg/l vykazovaly přijatelné růstové hodnoty. Ovšem při koncentraci 0,46 mg P/l, která je dvakrát vyšší než současná koncentrace P v rybníkách, vykazovali sinice mnohem vyššího růstové hodnoty. A dokonce při koncentraci 15 mg P/l dosáhly sinice nejvyšších růstových hodnot. Dá se tedy očekávat, že pokud bude koncentrace P v rybnících stoupat až k hodnotách 15 mg P/l bude v rybníce probíhat rychlý rozvoj sinic.
Na závěr experimentu jsem měla měřit růstovou rychlost v gradientu osvětlení, to jsem ale vzhledem k vyhlášení uzavření vysokých škol kvůli viru COVID-19 nemohla uskutečnit.

The bachelor thesis deals with the determination of cyanobacterial growth from ponds. The results are used to determine the growth potential in connection with eutrophication, i.e. increasing the concentrations of nutrients in water. Cultured strains of Cyanobium gracile A1 and Synechocystis sp. PCC7509 were used in the experiment. To determine their growth, static cultivation was used in culture plates with different well volumes of Thermo Scientific NunclonTM Delta Surface 96 Well and Cellstar 48 Well Cell Culture Plate, which were evaluated as the most suitable from the first part of the experiment. Cyanobacteria were cultured in concentrations from 0.06 to 30 mg P mg/l in Z medium. To measure the growth, a spectrophotometer measuring fluorescence and absorbance was set up at OD750 nm wavelength. At this wavelength cyanobacteria have negligible absorption and therefore turbidity was used to measure biomass. Growth curves were created from individual measurements and are shown in the form of graphs.
The results show that the highest concentration used, 30 mg P/l, caused growth inhibition. Cyanobacteria in an environment with a concentration of 0.06 mg P/l showed acceptable growth values. However, at a concentration of 0.46 mg P/l, which is twice the current concentration of P in ponds, cyanobacteria showed much higher growth values. And even at a concentration of 15 mg P/l, cyanobacteria reached the highest growth values. It can therefore be expected that if the concentration of P in the ponds rises to the values of 15 mg P/l, a rapid development of cyanobacteria will take place in the pond.
At the end of the experiment, I was supposed to measure the growth rate in the lighting gradient, but due to the announcement of the closure of universities due to the COVID - 19 virus, I was not able to fulfil that.

Cílem práce je stanovit míru růstu netererotrofních sinic rybničního fytoplanktonu. Výsledky budou sloužit ke stanovení růstového potenciálu sinic v rybnících a studentka se jejich pomocí pokusí vysvětlit pozorovanou abundanci sinic v jarním a letním období. Pro růstové experimenty budou použity kmeny rodů Planktothrix, Arthrospira, Microcystis nebo Cyanobium. Na již kultivovaných kmenech bude studentka měřit přírůstek biomasy za různých podmínek. Zaměří se primárně na rychlost růstu za různých kultivačních teplot, gradientu koncentrace fosforu v médiu a v gradientu osvětlení. Alternativně bude měřit primární produkci jako produkci kyslíku při různých teplotách a konstantního osvětlení. Z naměřených hodnot absorbance a hmotnosti sušiny studovaných kmenů budou sestrojeny růstové křivky a stanovena světelná a teplotní optima pro studované dominantní druhy vodních květů.
Získané výsledky pak studentka dále vyhodnotí pomocí počítačového SW (např. Statistica, Surfer, MS Office) a srovná s informacemi z dostupné literatury.
Výsledky pomohou vytvořit doporučení pro další management rybníků, např. aeraci nebo dokrmování rybí obsádky.

Studentka Petra Syslová seznámila komisi se svou bakalářskou prací. Tajemník komise, doc. Ing. Martin Kocour, Ph.D., seznámil komisi s posudkem vedoucího a oponenta bakalářské práce. Následně studentka zodpověděla doplňující otázky. Proběhla diskuse na téma práce. Studentka adekvátně a povětšinou správně reagovala na vznesené dotazy členů komise. Komise se při hodnocení vlastní práce ztotožnila s návrhem oponenta práce s tím, že v diskusi práce měly být i lépe diskutovány výsledky s ohledem k praktickému využití získaných poznatků.
